This guide assumes you are decently far into the game. If you have already killed specific people that I have mentioned in my Step by Step Guide you can just follow a specific challenge guide above. It is recommended you save before this so that you do not kill a lot of people on your main save.
Unless otherwise specified, these challenges all seem to register with any weapon of that type, not just GRA weapons.
General Tips and knowledge:
-- You do NOT have to do another playthrough. There are 3 of each challenge level that do not require killing main characters
Curios and Relics - 0/10000 - Cause damage with unique Mojave Wasteland weapons.
Master of the Arsenal - 0/10000 - Do damage with Gun Runners' Arsenal (GRA) Weapons.
You can knock both of these out by just causing 10,000 damage with a unique GRA weapon. My suggestion is to have a couple and get it over the course of doing other challenges and doing quests, or just go to the Thorn.
ONE STAR CHALLENGES
A Slave Obeys -- self explanatory. Nephi's Driver is held by Nephi who is in a Fiend camp north of the Samson Rock Crushing Plant. If you already have White Line Nightmare done there is no point for you to go all that way though.
Ne Ne Ne Ne -- Use any shotgun in V.A.T.S and go for the right arm
Overkill --
Benefit Or a Hazard -- go to Sunset Sasparilla Headquarters and kill as many as possible. If you need more go to REPCONN headquarters. Will test which one is better later
They don't belong in this world! -- Aliens, Centaurs, Deathclaws, Ghost people (Dead Money), Mr. House, Nightstalkers, Spore carriers, Spore plants, Think ** (Old World Blues)
The Same Could Be Said of All Religious Weapons -- Ghouls are pretty frequent. Head to Vault 34 for a lot of them
TWO STAR CHALLENGES
White Line Nightmare -- Usually in numbers and are hard kills
Fiends: north of the Samson Rock Crushing Plant (?)
Jackals: Nipton Road Pit Stop (4), Nevada Highway Patrol Station (4)
Vipers: Top of the hill next to Nipton (2), Down the hill towards Novac (3) but you have to be quick before the NCR takes them out
Dyin' Ain't Much of a Living -- To get a hit squad after you is as easy as pissing off the entire faction through story choices, mass murders, etc. Here are the type of weapons that the Cowboy Perk affects: dynamite, lever-action firearms, hatchets, knives, and revolvers.
Historical Propriety -- Self Explanatory. Caesar is in The Fort. Good luck getting out alive if you kill him. My suggestion is that if you are an enemy of the Legion, run into Caesar's tent, then run out. Then as his minions run out of the tent you can kill them one by one. Still tough thought
Even a God-King Can Bleed -- Goes perfectly with the one above. Bring Throwing Spears to cripple his head, then finish him off with a Knife.
Nyah! See? -- Killing spree on the Strip. Should not be hard to get the kills, just staying alive afterwards is the issue. Or, if you have not done the Beyond the Beef quest yet that is a great opportunity.
Talk About Owned -- Find Benny in The Tops and pickpocket his gun Maria, then cap him
Sic Semper Tyrannis -- Self Explanatory. President Kimball appears at the Hoover Dam to give a speech during a certain story quest.
Vault 13's Revenge -- Jacobstown is the best bet for heartless players. If you are not heartless go here:

Armed For Bear -- Just go to any place with an NCR presence and find the leaders. Ranger Stations are easier, but spread out.

-- Charlie: Comm Officer Stepinac, Ranger Beaumont (sometimes walks by on patrol, but never inside)

-- Foxtrot: Comm Officer Lenk, Ranger Kudlow
-- Mojave Outpost: (3) Major Knight, Ranger Jackson, Ranger Ghost

-- Camp Forlorn Hope
-- Camp Golf
Deathclaw Pro Hunter -- Go to Gypsum Train Yard. You can also fight Deathclaws in the Thorn if you complete the long quest, Bleed Me Dry.

Locations:
--Go to the old Nuclear Test Site and head east from there. You will find about 10 robots guarding a downed Vertibird.
--If you still need more go to Nellis airforce base, and those enemies respawn
Gun Locations: Here I will list the locations of where I am getting GRA weapons. You know they are GRA guns because they have (GRA) after them in the menu.
Mojave Outpost -- Lacey
Novac -- Cliff Briscoe

basically this part of the guide will highlight where certain challenges can be combined to be very efficient. I will also add an extra challenges for each level in case you have killed important people. You will complete these challenges if you follow this:

1. Go north of the Samson Rock Crushing Plant and south of the South Vegas Ruins East Entrance to find Driver Nephi and a handful of fiends. Kill them with the weapons specified in White Line Nightmare to get 5-7 points towards that. Use a Sawed Off Shotgun and aim for right arms until you get that challenge. Then take Nephi's Driver.
(6ish/20 White Line Nightmare, Ne Ne Ne Ne... complete)
2. Search around the area you are in for more fiends
(White Line Nightmare complete or close to complete)
If you still need a few kills for White Line Nightmare go to the Nevada Highway Patrol Station to find 4-5 Jackals, or Nipton Road Pit Stop for 4 Jackals
3. Kill Mr. House with Nephi's Driver
(A Slave Obeys Complete)
4. Go find Benny in The Tops and pickpocket his gun Maria, then kill him with it
(Talk about owned complete)
5. Go to REPCONN Test Site and use Maria to kill all the ghouls. In the Basement you will find a handful of Nightkin.
(The Same could be said of all Religious Weapons near complete)
6. If you need more Super Mutants, swing by Black Rock Cave for 3-4 Nightkin.
(Vault 13's Revenge complete)
7. Go East of the Old Nuclear Test Site to find 5 Mr. Gutsys and 5 Sentry Bots. Also there are 4 ghouls here that respawn every few days at the Test Site if you need more.
(Man-Machine Interface Complete, The Same could be said of all Religious Weapons Complete)
8. Go to the Gypsum Train Yard. There are plenty of Deathclaws here to kill. I would suggest using a stronger weapon to weaken all of them, then finish them off with the required weapons.

(Save for last because you can save before your massive killing rampage)
9. See above under Armed For Bear for all the locations of named NCR. The Ranger Stations are easier in my opinion because you only have to deal with 3-4 people at a time. Camp McCarran is not for the feint of heart, but will give you a ton of named NCR kills. All of the outposts, plus Mojave Outpost, and the one named NCR in Novac should be enough.
Extra
1. Go to REPCONN Headquarters and kill the Robots there.

Clean-up (places where enemies respawn if you have cleaned out the previous spots)
